Abstract
In conclusion, the cecum in the rat is the major site of fermentation by microbiota. Since the colon is right next to the cecum, colonic NQO1 induction as well as cecal inflammation indicate that cecal microbiota may play an important role in the hydrolysis to bioactive SF from GRP. However, the major detectable hydrolytic product by single Lactobacillus spp. evaluated in our study was nitrile, instead of isothiocyanate. This caused by decreased pH in media, known to favor nitrile formation, since microbiota in buffer did not alter this. The technical limitation of in vitro incubation studies may be that bacteria are sensitive to environmental changes not seen in vivo. Using in situ exposure to overcome this, we were able to directly monitor the hydrolysis of GRP, since the change for microbiota is the minimal. Bioactive SF was identified in the portal blood following introduction of GRP into the cecum.
