Abstract
Further characterization of SAS4 and SAS5 demonstrates that they define a new SIR-dependent silencing pathway in yeast. SAS4 and SAS5 are required for silencing at teloments while SIR1 is not. Also, SAS4 and SAS5 have opposite phenotype from SIR1 at HMR. These results indicate that SAS4 and SAS5 can function on a SIR1-independent silencing pathway. Furthermore, SAS4 and SAS5 contribute to silencing at HML. Double mutant analysis of silencing at HML between SAS4, SAS5 and SIR1 suggests that SAS4 and SAS5 may have similar function as SIR1 acting in establishment of silencing. Taken together, these evidence demonstrates that there exist multiple SIR-dependent pathways of silencing in yeast, which provide first genetic evidence of existence of multiple pathways of positon effects in any eukaryote.
