Abstract
In the second study, five carrot DHL lines were studied in order to identify the mechanism underlying glyphosate resistance. Uptake experiments, using radiolabelled glyphosate, showed higher or similar absorption levels in the resistant lines compared to the unselected control. Such results exclude uptake as a mechanism of glyphosate resistance. Inhibition studies of EPSPS by glyphosate however showed higher Ki values in three lines. One of these three lines (DHL 25), did not show consistent Ki values, but the observed values varied depending upon the growth stage of the cells at the time of each measurement, suggesting the presence of a less sensitive EPSPS enzyme that fluctuates in amount over time in this line.
