Abstract
"In order to characterize this secondary-binding interaction, a novel surface plasmon resonance technique termed ""analyte gradient-surface plasmon resonance"" (AG-SPR) has been developed and is described here. AG-SPR can be used to rapidly characterize macro-molecular interactions without requiring surface regeneration and has been used to monitor the binding of two truncated P450s with a phosphatidylcholine supported lipid bilayer. The data has been fit to a kinetic model that incorporates a semi-empirical available surface function to account for the effect of steric interactions between pre-adsorbed molecules. The results indicate that P450 does in fact contain a peripheral binding interaction. However, in order to determine an accurate binding affinity, additional factors must be incorporated into the kinetic model."
