Abstract
We provided the evidence that both microtubules and actin filaments are required for the proper localization of FMRP-RNP particles. Furthermore, FMRP associates with both microtubule- and actin-based motors, kinesin-1 and myosin Va. In order to unambiguously identify the detailed molecular mechanisms for RNA transport, we developed an in vivo functional assay and went on to provide evidence that dFMRP and associated mRNAs are actively transported by molecular motors. We were able to not only identify the motors that are required for the movement of dFMRP-RNP, but also look at the roles of conventional adaptors for the respective motors. Interestingly, both opposite-polarity motors, kinesin-1 and cytoplasmic dynein, are required for dFMRP-RNP transport, implying a coordination model rather than tug-of-war model for motor activities. Furthermore, while KLC is dispensable, dynactin is required for dFMRP-RNP transport, indicating a novel attachment of kinesin-1 to dFMRP-RNP.
