Abstract
Satellite Tobacco Necrosis Virus RNA (STNV RNA) is an efficient, eukaryotic mRNA lacking a 5$\sp\prime$-terminal cap group or 5$\sp\prime$-terminal covalently-linked protein. Addition of a 5$\sp\prime$-terminal cap group does not enhance the translation of STNV RNA. This suggests that STNV RNA contains some feature(s) which compensates for its lack of a 5$\sp\prime$-terminal cap group. Interestingly, the 5$\sp\prime$-untranslated region of STNV RNA contains a nucleotide sequence that is complementary to the 3$\sp\prime$-terminal region of the 18S rRNA.
Mutant transcript STNV RNAs that lack the potential to hybridize to the 3$\sp\prime$-terminal region of the 18S rRNA, lack cap-independence. Additionally, mutant transcript STNV RNAs that lack either the potential to hybridize to the 18S rRNA, or the natural AUG translation initiation codon initiate translation faithfully from the natural translation initiation site. Mutant transcript STNV RNAs that lack both the potential to hybridize to the 18S rRNA and the natural AUG translation initiation codon do not initiate translation efficiently or faithfully.
Further, STNV RNA contains a unique 5$\sp\prime$-terminal secondary structure. Mutant transcript STNV RNAs lacking this 5$\sp\prime$-terminal secondary structure do not have altered translational efficiency, cap-dependency or specificity indicating that the unique 5$\sp\prime$-terminal secondary structure of STNV RNA does not play an integral role in its cap-independent translation. These data indicate that a STNV RNA:18S rRNA hybridization functions in the cap-independent initiation of STNV RNA translation.
