Abstract
Onion seeds had cracks, and fungal infection was found over and inside the hilum and seed cracks. During storage dehydration and fungal infection may lead to membrane damage. Aged seeds had a high electrolyte leakage and encountered hydration stress which reduced seed germination and seedling vigor.
Tissue culture may be a useful tool to contribute to seed aging problems. Multiple shoots were obtained from seedling shoots, onion-set shoot tips, onion bulb shoot tips, and inflorescence. Shoots produced can be induced to form bulbs and can be transferred to the soil without acclimatization.
Artificial sets were obtained from regenerated shoots or directly from inflorescence in a medium with high sugar concentration. The production of artificial sets may overcome the problem of cost in regenerating in vitro plants; facilitate selection, handling and storage; and be useful in the production of disease-free germplasm.
