Abstract
Sweet corn genotypes containing the shrunken2 (sh2) endosperm mutation are associated with reduced emergence and poor seedling vigor. Shrunken2 genotypes with known differences for field emergence and vigor were used to investigate the contribution of seedling cell division to emergence and seedling growth. Variation among genotypes was observed for all of the cell division cycle phases which include G1, S, and G2. The S phase, when cells are undergoing DNA replication, showed the closest association to seedling growth. Inbred lines homozygous for starchy (Su1) endosperm displayed a significantly higher proportion of nuclei in the S phase than the same inbred homozygous for the sh2 endosperm mutation. Among all the genotypes tested, the proportion of nuclei in the S phase was positively correlated with field emergence and seedling vigor.
Variation was observed among sh2 genotypes for the rate of cell division in seedling shoots, which was measured by a method using nitrogen-15 to label DNA. Rates of DNA replication during the S phase in six to nine day old sh2 sweet corn seedlings were highly and positively correlated with field emergence and seedling growth rates.
