- The ability to properly delineate fungal species is crucial in understanding species level ecological roles and understanding species level community diversity.This study evaluates the effectiveness of sequence similarity (at 97% and 99%), the GMYC model, and the bPTP model to delineate 51 species within the Penicillium section Aspergilloides, and evaluats the effectiveness of these models to correctly delineate 13 unknown Penicillium species. DNA extraction, PCR amplification, and sequencing was conducted on the 13 previously delineated Penicillium isolates to determine their identity based on multigene phylogeny. In this study, we provide insight into the limited capabilities of bPTP, GMYC and sequence similarity in delineating Penicillium species. Between the two tree based models, bPTP gave a wider CI, however, it was more accurate than the GMYC model. The GMYC model performed the poorest in delineating the 13 unknowns as well as the species in the mock community.We recommend the use of both sequence similarity at the 99% threshold and the bPTP model, as they have generally performed well in delineating species
